Acridine orange is a cell-permeable, cationic fluorochrome dye widely utilized in biotechnology and molecular biology for the differential staining and visualization of nucleic acids. When intercalated into the double-stranded helix of DNA, the monomeric dye emits a bright green fluorescence at approximately 525 nm, whereas its electrostatic interaction and subsequent aggregation along the phosphate backbone of single-stranded RNA results in a metachromatic shift to red fluorescence at roughly 650 nm.
