TAE Buffer 10X is a concentrated solution composed of Tris base, acetic acid, and EDTA, commonly used for the electrophoresis of nucleic acids. It functions as both a running buffer and a component in agarose gels, maintaining a slightly alkaline pH (typically 8.2–8.4) to protect DNA from hydrolysis while providing ions for electrical current. The presence of EDTA in the buffer sequesters divalent cations like magnesium, thereby inactivating DNA-degrading nucleases during experimental procedures.
